| Rat monoclonal antibodies We make monoclonal antibodies to protein antigens or fusion proteins. For customers who do not have ready to use antigens, we provide antigen preparation services that include cDNA expression, peptide design, synthesis and cross-linking peptides onto carrier proteins.
Producing monoclonal antibodies normally takes 20-24
weeks, which can be divided into three stages as below.
(1) Immunisation of 5 rats and serum ELISA after
2nd and 3rd injection. Usually antiserum will show positive when
it is diluted 50,000 times or more. If antibody titre is not satisfactory,
we shall do one more injection.
(2) Fusion and ELISA screening. We usually screen
1400 cells and can identify 10 to over 100 positives. If customers
want to further select cells themselves with their assays, we can
collect about 300 microliter supernatants from all ELISA positive
cells and send them to customers.
(3) Subcloning, expansion and freezing down cells.
Customers will decide which clones they want to keep and we do not
limit cell numbers to be subcloned. We do subcloning until all cells
are positive. At the end, we shall send cells in culture to customers
and freeze down two vials as backup. Unless specially arranged,
we shall discard cells 6 weeks after customers have received cells.
On request by customers, we shall do antibody further
purification and characterisation.
We charge $4200 US as dissected below for the first
cell line and $50 US for each of the additional cell lines. Customers
can choose to pay before each work stage or pay the full amount
upfront. Customers can terminate a project before any stage. We
will refund customers upfront payments made for any work that has
not been initiated.
